Cryo-Ultramicrotomy in Biology

Abstract:Cryo-ultramicrotomy gains increasing interest in the life science community. The sample preparation by plunge freezing or high pressure freezing, cryoultramicrotomy, cryo-transfer and cryo-TEM preserve living matter close to the native state. The cutting process is accompanied by a number of artefacts and problems. A crucial part in cryo-sectioning is the section pick-up. The transfer of the sections onto the grid is difficult and new micromanipulators for the handling of the section ribbons, which make it easier and less depending on the operator's skill, will be demonstrated. Possible scenarios for reducing these artefacts will be discussed.
